چکیده

The ventricular zone (VZ) of the embryonic dorsal telencephalon is a major site for generating cortical projection neurons. The transcription factor Pax6 is expressed in apical progenitors (APs) residing in the VZ in a rostrolateral-high to caudomediallow gradient and its levels are highest at early stages of corticogenesis. Previous studies mainly focused on Pax6 / mice have implicated Pax6 in cortical progenitor proliferation, migration, neuronal differentiation and formation of superficial cortical layers. We wish to explore the developmental significance of Pax6 expression levels in cortical progenitors at different developmental stages and levels across the rostral-caudal axis of the cortex. In order to address this issue, we are using a floxed Pax6 allele, where the exposure to Cre recombinase results in excision of Pax6 and initiation of YFP expression. Use of a tamoxifen-inducible Emx1-CreER driver allows us to remove the gene from cortical progenitors in a mosaic manner at times coincident with deepor superficial layer neurons being generated. Our preliminary results suggest that when Pax6 is removed from cortical progenitors at mid-stages of corticogenesis, the thickness of the postnatal cortex is severely reduced due to a remarkable under-production of superficial layer neurons. Deletion of Pax6 at late stages of corticogenesis results in the contribution of late-born Pax6 null cells to deeper laminar positions compared to Pax6 cells. This suggests that the normal low expression levels of Pax6 in late APs might have an effect on the specification of superficial layer neurons.

Actin-binding proteins.

Actin is an essential component of the cytoskeleton and plays a crucial role in eukaryotic cells. The actin cytoskeleton functions in the generation and maintenance of cell morphology and polarity, in endocytosis and intracellular trafficking, in contractility, motility and cell division. In cells, the assembly and disassembly of actin filaments, and also their organisation into functional high...

Cdc28–Cln3 phosphorylation of Sla1 regulates actin patch dynamics in different modes of fungal growth

A dynamic balance between targeted transport and endocytosis is critical for polarized cell growth. However, how actin-mediated endocytosis is regulated in different growth modes remains unclear. Here we report differential regulation of cortical actin patch dynamics between the yeast and hyphal growth in Candida albicans. The mechanism involves phosphoregulation of the endocytic protein Sla1 b...

Plasma membrane restricted RhoGEF activity is sufficient for RhoA-mediated actin polymerization

The small GTPase RhoA is involved in cell morphology and migration. RhoA activity is tightly regulated in time and space and depends on guanine exchange factors (GEFs). However, the kinetics and subcellular localization of GEF activity towards RhoA are poorly defined. To study the mechanism underlying the spatiotemporal control of RhoA activity by GEFs, we performed single cell imaging with an ...

Verprolin: a cool set of actin-binding sites and some very HOT prolines.

Spatiotemporal organisation of eukaryotic cells is established and maintained by the cytoskeleton, a highly dynamic and complex network of structural and signalling proteins. Many components of the cytoskeleton are functionally and structurally conserved between humans and yeast. Among these are verprolin (Vrp1p) in yeast and its human ortholog Wiskott-Aldrich syndrome protein (WASP)-interactin...

The Effect of Resistance Training and Endothelial Stem Cell Injection on Βeta-Actin, Phosphorylated and Total AKT of Skeletal Muscle in Type 1 Diabetic Rats

Background: Type 1 diabetes is associated with decreased skeletal muscle capillary and improper regulation of angiogenesis pathways in skeletal muscle. This research intended to study the effect of resistance training and endothelial stem cell injection on βeta-actin, phosphorylated and total AKT of skeletal muscle in type 1 diabetic rats. Methods: In this experimental study, 36 male Wistar ra...
